Evolutionary conservation and versatility of a new set of primers for amplifying the ribosomal internal transcribed spacer regions in insects and other invertebrates

چکیده
The evolutionary conservation and versatility of a new set of nuclear primers for the amplification of the ribosomal internal transcribed spacer regions in insects and other invertebrates have been studied. These primers, conserved across Insecta and other invertebrates, are based on a comprehensive taxonomic survey of the current DNA sequence databases. Their versatility was demonstrated by extensive polymerase chain reaction assays in 16 species from two arachnid orders, eight insect orders, three invertebrate and vertebrate chordate orders and by direct sequencing of the amplified products. The availability of these primers to the insect research community should facilitate the use of internal transcribed spacer regions in intraspecific studies as well as phylogenetic analysis of closely related taxa.

Study on coprophilous fungi: new records for Iran mycobiota
In a study on coprophilous fungi, different samples including cow, sheep and horse dung and mouse feces were collected from different locations in West and East Azarbaijan provinces (NW Iran). Isolation of the fungi was done based on moist chamber culture method. Purification of the isolated fungi was done by single spore culture method. Several fungal taxa were obtained. Identification of the ...
متن کاملSpecific Primers For Identification and Detection of Three Major Desert Truffle Species in Iran with Special Reference to Terfezia claveryi
ABSTRACT- Desert truffles are hypogeous ascomyceteous ectomycorrhizal fungi, occurring in arid and semi-arid ecosystems. A PCR-based method was developed for the identification of 3 major desert truffles of Iran: Terfezia claveryi, Tirmania pinoyi and Tirmania nivea based on internal transcribed spacers of rDNA. Two specific PCR primers were designed for T. claveryi, 4 for T. pinoyi, and 2 for ...
متن کاملFomitiporia mediterranea, a new basidiomycete species for mycobiota of Iran
During last decade, the decline of elm trees was observed in Fars province (Iran) due to a wood decaying agent in heartwood of the trees. Unknown basidiomycete isolates were frequently isolated from such trees. The objective of the present study was to identify the basidiomycete isolates by molecular phylogenetic analysis of the internal transcribed spacers of ribosomal DNA. For this purpose, i...
متن کاملMolecular phylogeny of the family Araceae as inferred from the nuclear ribosomal ITS data
The internal transcribed spacer regions of nuclear ribosomal DNA are widely used to infer phylogenetic relationships in plants. In this study, it was obtained the ITS sequences from 24 samples of Araceae in Iran, representing 3 genera: Arum L., Biarum Schott. and Eminium (Blume) Schott. Phylogenetic analyses were conducted by Bayesian inference and maximum Parsimony methods. Cladistic analysis ...
متن کاملComparative Phylogenetic Perspectives on the Evolutionary Relationships in the Brine Shrimp Artemia Leach, 1819 (Crustacea: Anostraca) Based on Secondary Structure of ITS1 Gene
This is the first study on phylogenetic relationships in the genus Artemia Leach, 1819 using the pattern and sequence of secondary structures of internal transcribed spacer 1 (ITS1). Significant intraspecific variation in the secondary structure of ITS1 rRNA was found in Artemia tibetiana. In the phylogenetic tree based on joined primary and secondary structure sequences, Artemia urmiana and pa...
